A new water distribution line was run down my street, replacing the 80 yo original. I received notice my house will be switched from the old to the new line tomorrow. They will be cutting the supply line on the house side of the meter.

So when they do that, I assume the bulk of my house lines will drain. Thus when reconnected and I turn on a faucet for the first there will be a ton of air in the lines.

Does it make sense to turn off my main shut off in the basement, thereby isolating the house pipes?

Once the pressure in the lines is released, it should airlock and not drain a whole lot if all of the faucets are closed. I’m guessing they’re not gonna bleed the new line all the way to your house so you’ll probably still have some air work it’s way through.
